Should I use AVIF instead?

Should I still keep a PNG fallback?
No. Every browser released since 2020 supports WebP, including Safari 14 and later. A fallback is now dead weight in your markup.
Does converting to WebP lose transparency?
No. WebP supports a full alpha channel, which is what makes it a real replacement for PNG rather than for JPEG only.
